The short answer is yes you can KO someone with one punch in MMA, and the strikes you learn in Muay Thai training and Western boxing are the best for the job, however, for this to be accomplished there are several factors that must be present:
1) Clear target: For the quickest and most effective KO's in MMA the best target is the head. This must be hit so that the brain is jolted violently within the skull. We must train so that when a target presents itself we can strike with speed, follow-through and power, as soon as the opening occurs and before the opponent can react to defend the hit. 2) Timing must be right: The best time is when the opponent has committed themselves to moving in a particular direction and you catch them with a shot that either strikes from the opposite direction or from an angle that rotates the head such as the hook or elbow. You must train this constantly as timing is one of the earliest attributes that we lose when we are not training
3) Your balance and positioning: You must have yourself positioned so that you are well balanced and able to use the correct body mechanics to strike with, particularly in MMA where you are always in danger of being taken down.
This will ensure that you hit with maximum power and follow-through. Your positioning must also take into account your opponents attack and make it difficult for the opponent to defend cover or counter-attack should your strike fail.
4) Selection of the right strike. The best strikes for scoring a KO in MMA are the Muay Thai and Western boxing striking tactics. This is a result of several factors:
a. These strikes are used in full contact fighting matches and therefore have evolved to be effective KO punches in a rapidly changing fight environment.

c. In Muay Thai training and Boxing we are constantly training our strikes to KO our opponent.
The most useful strike for KO's in MMA are; Straight right (left for southpaws), Hook (both left and right, Uppercut (again both left and right), Elbow (several angles) and the knee which although it is not a punch I include because it is one of the best weapons to KO an opponent in MMA, when the opportunity presents itself (opponent over-commits head in the shoot).
Thus we can conclude that it is most certainly possible to KO someone in MMA with one punch and there are plenty of examples of this occurring. In addition, one sure way that you can increase your ability to do this is to include Muay Thai training in your MMA game.
